Hi @vhhbk59, I'm sorry to hear that.
Have you tried inserting a SIM in slot 2 without having one in slot 1?

I flashed back to build 26.1.B.2.102 (android 5.0), 2 SIMcard slots worked normally. Then I tried to update via OTA, but after the first update to 26.1.B.2.129 (still android 5.0), the SIM2 slot didn't work anymore. Now I'm desperated.

When your phone is updated to the latest available software version, could you take a bug report and send it to me? I'll then forward it for investigation.
To do this:
- Go to Settings > About phone > Build number, tap 7 times on the build number.
- Go back and select Developer options then turn on "USB debugging".
- Select Logger buffer size and change it to "4 M".
- Restart your phone, make sure that you have two SIM cards inserted.
- Go back to Settings > Developer options, select "Take bug report".
When the report is ready you will receive a notification. Tap the notification to share it. I suggest you upload it to Google Drive and then choose to share a public link to the upload and include the link in the PM. Please also include your device's IMEI.
